The church was built as part of the expansion of the castle complex under Duke (later Grand Duke) Carl II of Mecklenburg-Strelitz (1741–1816). Hohenzieritz Castle served him as a summer residence and was developed into a prestigious estate from the 1770s onwards, including an English landscape park (one of the oldest on the continent).

The Evangelical Lutheran Church in Prillwitz is a remarkable example of regional architectural history. The original nave dates back to the Middle Ages, while in the 18th century a half-timbered church was built, presumably on the foundations of the previous building.
